My issue is that the show spends so much time on the quirky Whedon comedy, Millenialogue, and Simpsons esque B-plots that by the time a scene like that happens it feels totally unearned and doesn't register for me because at that point the characters are just obnoxious. Their problems are tedious. Their (totally self inflicted) misery is comical.
Hell, even that clip you posted does this exact thing: Bojack has a serious moment of introspection and raw emotional vulnerability where he levels a very pointed and weighty cry for help against someone that he respects, and then some ugly fatass with an obnoxious voice stands up and kneecaps the entire scene by reiterating one of the series' longest running jokes.
