Really, nothing's in motion at this point. Joaquin isn't even confirmed and it's more likely imho that they're going to look into making these one-off movies about the other DC villains, which I think would be a much better direction to take. Director Todd Phillips will also be developing at least one DC origin story in the mould of the Clown Prince of Crime's billion-dollar hit.
As revealed by sources close to The Hollywood Reporter , Phillips is in talks to direct the Joker sequel. He would also potentially write the script alongside Scott Silver, who co-wrote the first Joker movie.
But what about Joaquin Phoenix? THR says Warner Bros. has an option to bring him back for a sequel.
Phillips may also be diving further into DC's universe. During an October meeting, he reportedly asked Warner Bros. for the rights to create a suite of origin movies starring DC's comic book characters. He didn't quite get that, but will supposedly be producing at least one origin story.
News of a Joker sequel has been a long time coming, even before the movie's October release.
Todd Phillips exclusively told sister publication Total Film back in August that he was open to doing a potential sequel, saying, "I would do anything with Joaquin, any day of the week. There’s nobody like him. If he was willing to do it, and if people show up to this movie, and Warners came to us and said, ‘You know what? If you guys could think of something…’ Well, I have a feeling that he and I could think of something pretty cool.”
He later said, he had "no plans for a sequel" but, with today's news, it's clear that whoever was pulling the strings had the last laugh: we're getting the Joker sequel we've been clamouring for ever since Arthur made his explosive late-night debut.

I don't think it'll be a direct sequel or that there'll even be one. It's pretty much the same stuff we've been hearing about a joker sequel from Phillips which is literally " yeah I've thought about it" and then saying he doesn't want to. But IF they were I would hope it wouldn't rehash the first movie like most sequels do, either way I have the feeling there might be a little more studio intervention if it's greenlit.Article for it

Making similar supervillain origin stories with a dark tone and small budget = good idea

You make a very strong point, but I have two options available to me in terms of likely lessons taken from a movie that made 15x its budget in pure, shiny profit:Is it? The second this kind of sincere, refreshing movie becomes part of an assembly line process is the second they will start to fail.
And you know the dummies who run these studios would want one for every Batman villain. But it doesn't get bigger than the Joker in the Batman universe. There are other great villains, but none as great as he is. Does anyone really want a dark, depressing Clayface movie? Poison Ivy? Killer Croc? I would love to see more nuanced, adult takes on these characters, sure. But I don't want them to ape this film to do it or create a "Joker formula."
I love this movie, and I really don't want to see it's strengths diluted with tons of bad (or even good!) imitations. (Though I couldn't be happier for Phillips that he's finally broken into dramatic films.)
To be fair, Killer Croc already has a dark depressing backstory.
Long and short of it is he was raised by an alcoholic aunt who abused him. He was also born with a skin condition that made him look reptilian, which caused him to be ostracized from other people. As time goes on, he becomes more animalistic.
Frankly, I think a movie based on Killer Croc would be way more heartbreaking than Joker, since you could make it all about the concept of abuse and what that does to people. Joker is already uncomfortable due to the way it tackles mental illness, but Killer Croc has potential in the way it could show how abuse can essentially damn someone to a life of crime.
I'd love to see a based Poison Ivy movie in which her sjw leanings, misandry, and overall shitty reclusive mentality is the cause of her own suffering. Ivy is my third-fave Batman Rogue because she's clearly a very unhappy lonely person that I feel for but know that it's 100% her own fault.

It'd be like making a sequel to Taxi Driver. Might be an unpopular opinion but I can't stand the idea of Harley with this Joker. You couldn't make her work in this world without completely gutting her character. FFS she was invented for a cartoon.
