Absolutely. If there's a way to cheat, John will have sniffed it out at the get-go. Like all trannies, John thinks rules (and things like etiquette, manners, societal norms, and simple human decency) are for the little people.

John's use of Photoshop's autotrace function was an inking pass, you monster! All artists use inking passes (despite the fact that inking passes -- like alpha blades -- exist only in John's con artist imagination).

Speaking of not bragging about meaningless accomplishments, there's a lot of runners that won't bother posting, let alone bragging, about anything that's not top 10. Maybe top 20, depending on how competitive/close the category is. Generally, when speedrunners do brag about records, they don't add the "in the world" bit like Wu constantly does as well, not that we need further proof Wu is just grandstanding and trying to come off as some kind of elite gamer.

I assumed it was something to do with SMB2 being a reskinned game from an unrelated franchise.

No, he's referring to Super Mario 3D World, which came out twenty-five years after SMB2, not twenty-six (amazing how he can fuck up even the simplest things). John only cares about this game because he gets to satisfy his autogynephilia by playing as Peach the whole way through. In further dumbassery on John's part, Nintendo did in fact announce 3D World would be getting a Switch port during the same Direct, but he can't help but spam his fetish at unrelated tweets.

I'm guessing this game doesn't count for whatever reason. Shouldn't Peach be able to stand on her own merits and be considered valid outside of a Mario game, John? Awful misogynistic of you.

His bragging about the RE3 speed run still cracks me up. Anything more than 45 minutes is incredibly slow - even an average player can nail the game in around an hour with the post campaign cheats Bri uses.
