Good question. Trying to think of some. Penicillin I suppose, potassium bromide for epilepsy (they thought it was caused by excess wanking, but the potassium bromide does actually have anti seizure activity.) I think benzos were found by accident looking for antibacterial drugs, spilling something and noticing that the mice or staff or whatever really chilled right out.
This is why they do stuff now like screening against whole libraries of molecules - Tim Haystead’s work on this led to HIV drugs.
However even if you find a drug and know it works you’ve still got to get it though the trial process, with various phases to show safety, efficacy etc. That takes the best part of a decade (does not apply to Covid vaccines ofc… and it won’t apply to whatever they want to Inject you with next.)
There’s a really good article waiting to be written to answer that question. ‘Lucky accidents in pharmacology.’
Viagra was in development as a blood pressure medication, the boners were a side effect they couldn't engineer out.

Imodium was an attempt to make a non-addictive opiate, but taking out the things that make opiates addictive removed its ability to work as a painkiller while keeping the side effect of constipation.

Not quite pharmacological, but many artificial sweeteners were found by mistake and some good old derring-do. So many of them were discovered because a chemist decided to taste what they made.

Chemists are weird.

A lot of erectile dysfunction is essentially an issue of microvessels in the penis and is a symptom of cardiovascular issues. Viagra became popular because people have gotten unhealthier and it's fucking up their T levels (unhealthy diet and fat) and the vascular health in their penises.

Side note, saw a tweet that said ICE is "snatching up Somalis left and right" right now which gave me a good chuckle.
